Three men were arrested in Reeston, near East London, on Monday for the possession of unlicensed firearms, Eastern Cape police said.
Police were following up on a tip-off, spokesman Captain Mluleki Mbi said in a statement.
"On seeing the police, the suspects fled on foot and the police gave chase," he said.
One man was injured during an exchange of bullets with police.
Mbi said the three men were aged between 20 and 21 and charged with the possession of unlicensed firearms.
"One 9mm pistol and a handmade gun were recovered from the suspects," he said.
They would appear in the East London Magistrate's Court soon.
